Transgenic/Knock-out Mouse - Equipment
The Transgenic/Knock-out Mouse Shared Resource possesses all necessary equipment for the generation of transgenic and gene targeted mice that are located within the mouse facility or laboratory as appropriate. Within the barrier facility of the Child Health Institute of New Jersey (CHI), TG/KO has two procedure rooms containing two state-of-the-art microinjection systems capable of both pronuclear as well as blastocyst injection. This system includes Nikon TE300 and TE2000S inverted microscopes equipped with DIC optics, two sets of Eppendorf Transferman NK micromanipulators, Eppendorf Celltram and an Eppendorf Femtojet injection controller and a PrimeTech Piezo-drill. Additional equipment includes Leica S8-APO stereo microscopes for embryo handling and re-implantation, a CCD camera and color monitor for training, a Napco CO2 incubator for embryo culture, a deFonbrune microforge, a Sutter Pipet puller, and a Bio-Cool III controlled cooler for embryo cryopreservation.
Within the CHI building, the resource has a laboratory and tissue culture area that is well equipped for standard molecular biology tasks and for ES cell culture. This laboratory contains equipment for molecular biology experiments (MJ Research PCR machine, Hybaid HD1 oven, Bio-Rad GelDoc system, gel electrophoresis power supply and tanks, -20°C freezers, refrigerator, liquid nitrogen storage tanks, microcentrifuges, Stratagene 2400 UV- crosslinker, Eppendorf 5810R centrifuge and a -80°C REVCO etc.) and an area for tissue culture (equipped with a tissue culture hood, Napco CO2 incubator, Biosafety cabinet, table-top centrifuge and Nikon TS100 inverted microscope, Bio-Rad Xcell GenePulsar electroporator, etc).
